Because you just cant enough webpages devoted to this life saving tool, heres another link to this great resource!

I would only add the following modifications to their guide:

First: when you download ComboFix, rename it! I usually go with “CF1.exe” but “bananarammajamma.exe” works just as well.

Second: If possible, run ComboFix in Safe Mode with Networking. This increases the chances of thorough cleanup and reduces the risk of a failed run.

Lastly: The Guide says nothing about ComboFix rebooting Windows. It will do this if the infection is nasty enough. Even though ComboFix is rebooting Windows, make sure it reboots back to Safe Mode. That way you can run MalwareBytes and SpyBot after its done.
